Embryo spent culture medium has been intensively investigated, considering its promising feature for non-invasive bioanalytical techniques in in-vitro fertilization (IVF). Despite, isolating DNA from such samples is quite challenging due to its small volume. Carrier RNA is reported to exhibit DNA retrieval effects and commonly employed in various limited biological samples, but there are no reports regarding its benefit on embryo media. Therefore, we aim to evaluate the competence of cRNA on isolated DNA from embryo medium and analyzed its optimal volume as there are also no records respecting its ideal volume to obtain decent outcomes. Results showed that cRNA significantly increases DNA amounts in the cRNA treated group (p<0.001), but the D-4/D-5 medium yielded similar (p=0.684). Pearson test demonstrated no correlation between cRNA volume vs. total retrieved DNA (r=0.760, p=0.80), and Whole Genome Amplification (WGA) was shown to increase DNA in the treated group (p=0.022), but not in the untreated group (p=0.128). Additionally, electrophoresis successfully resulting in a thick and thin band of TH01 locus signifies the cRNA competence. In conclusion, our study suggests that cRNA addition is essential in embryo medium extraction as it increases initial DNA that crucial for downstream application. However, the optimal volume could not be determined in the current study since the initial amount of DNA in the medium is unknown. BACKGROUND: Renal tumor biopsy is beneficial as it is capable of distinguishing between histological types of renal tumor; hence, it plays an important role in deciding the best therapy regimen. AIM: This study aims to evaluate the clinical experiences of renal biopsy in Cipto Mangunkusumo National Referral Hospital (RSCM), with both a percutaneous and open approach. It also aims to analyze the indications, results, intraoperative information, and complications of the two approaches. METHODS: This study was conducted using the retrospective cohort design; meanwhile, data were collected from RSCM from 1990 to 2019. The biopsy sample was taken using percutaneous and open renal biopsy, while comparative analysis was done between the two biopsy approaches. RESULTS: Data were collected from 33 patients that underwent renal biopsy from 1990 to 2019. Majority of the cases were diagnosed as unresectable renal tumor, while histological examination found clear cell carcinoma in most of the cases (73%). Furthermore, the open approach showed longer duration and higher blood loss compared to percutaneous technique with median 60 (30–120) versus 30 (5–60) min (p < 0.001) and 100 (5–650) versus 2 (1–5) ml (p < 0.001), respectively. In general, complications were reported to be low for both techniques. CONCLUSION: Based on the results, percutaneous renal biopsy has similar efficacy and complications rates in tumor sampling for histopathology together with open approach. In the present study, the potency of rice bran as an antioxidant was examined. Rice bran is a by-product of the rice milling process, despite being a rich nutrient, it has limitations in food application. In this study, we used carbon tetrachloride-induced rats (CCl4) as a model of oxidative stress and examined the effect of extract IPB 3S rice bran supplement (RBS) on testis and liver endogenous antioxidant. The testis and liver were used as the representative organs which prone to exposure to reactive oxygen species (ROS). We used 150 and 300 mg.kg-1 Body weight (BW) of RBS. The Concentration of glutathione (GSH) in both organs was measured. All groups administered by RBS had significantly higher GSH levels compared to the CCl4 group, both in testis and liver. The dose of 300 mg.kg-1 BW RBS had a significantly higher GSH level in testis, while 150 mg.kg-1 VA RBS had a significantly higher GSH level in the liver tissue compared to the control group accordingly. Thus, the rice bran supplement enhances GSH levels in rat’s liver and testis which potentially has protective effects. © 2021 Society of Pharmaceutical Sciences of Ankara (FABAD). Burkholderia are intracellular pathogenic bacteria which can produce biofilm. This biofilm protects the intracellular pathogenic bacteria from antibiotic treatment and the immunological system of the host. Therefore, this review aims to describe the capacity of Burkholderia to form a biofilm, the regulation of its biofilm formation, the efficacy of antibiotics to eradicate biofilm, and the novel therapy which targets its biofilm. Burkholderia's biofilm is characterized by its lipopolysaccharides, exopolysaccharides (EPSs), biofilm-associated proteins, and eDNA. Its regulation is made by quorum sensing, c-di-AMP, sRNA, and two component systems. Many antibiotics have been used as sole or mixture agents; however, they are not always effective in eradicating the biofilm-forming Burkholderia. Inhibitors of quorum sensing and other non-conventional antibiotic approaches are promising to discover effective treatment of Burkholderia infections. © 2021, Bentham Science Publishers. Introduction: Nasopharyngeal carcinoma (NPC) is a multifactorial disease with genetic, viral, environmental and lifestyle-related risk factors. Epstein-Barr virus (EBV) can promote the oncogenic transformation of an infected cell into malignant. EBV encodes many stimulating products including Epstein-Barr virus nuclear antigen-1 (EBNA-1) which plays a key role in the regulation of gene expression and replication of the genome in the latent period of infection. EBNA-1 in serum and tumour tissue of NPC patients correlates with NPC prognosis. Moreover, the presence of EBV DNA in serum samples from NPC patients' blood circulation can be used as an early marker in the diagnosis of NPC. Objective: The objective of this study was to find effective methods for monitoring the progress of NPC patients undergoing radiotherapy and therapeutic efficacy by observing the changes in EBV DNA in serum and saliva. Methodology: The pre-experimental design compared blood and saliva taken from a pretest and post-test group of NPC patients before and after radiation therapy. The concentration of EBV DNA was measured in the serum and saliva after amplification using quantitative polymerase chain reaction (qPCR) with compatible primers for the EBNA-1 gene. The data were statistically analysed by paired T-test. Results: Highly significant (p = 0.0001) increase in cycle threshold qPCR and decrease in the mean concentration of EBV DNA (p = 0.0001) were observed in serum samples, but no significant changes were observed in saliva. Conclusions: The results suggest that EBV DNA in serum can be used as the gold standard and a marker for monitoring the response to radiation therapy in NPC patients, whereas the examination of EBV DNA from saliva samples is not accurate and thus, not appropriate. © 2021 ecancer Global Foundation. Background: Management for male infertility can be difficult for some cases. Surgical intervention has long been thought as the last resort to help married couples to conceive. The current guideline recommends testicular sperm extraction with microsurgery technique (microTESE) in severe cases of male infertility. However, the success rate still varies. Thus, a new strategy was needed to further increase the sperm retrieval success rate. Case Presentation: A 39-year-old male with a history of failed sperm extraction, non-obstructive azoospermia (NOA) and Y-chromosomal microdeletion came to the fertility center to undergo sperm retrieval. Fine needle aspiration (FNA) Mapping was performed prior to microTESE to increase the accuracy of sperm retrieval. After further examination with laser assisted immotile sperm selection (LAISS), five spermatozoa were found. The increase in dementia incidence among the elderly is directly related to aging, which is associated with changes in bodily functions and other health-related risk factors. Alzheimer’s dementia is the most prevalent form of dementia, and individuals in the late stages are predominantly dependent on other family members. Therefore, it is important for families, as the closest support group, to recognize common symptoms early. Objective: To provide a family-friendly guide to the ten common symptoms of Alzheimer’s dementia. Methods: This is a descriptive survey-based research that included 354 families comprising elderly people (≥60 years) residing in Jakarta. The instrument aimed at identifying ten common Alzheimer’s dementia symptoms in Indonesia. Descriptive statistical analysis based on frequency tables was used. Results: The participant’s major characteristics were age ≥66 years (52.3%), female sex (70.3%) and primary school education (87.3%). The predominant symptoms experienced by 42.4% of the elderly included forgetting recent events and asking questions and narrating a particular detail repeatedly. The remaining 35.6% demonstrated signs of forgetting where an item was placed and frequently suspecting others of theft and concealment of personal items. Conclusion: The symptoms of frequently forgetting new events and the location of personal belongings are of particular concern for families, as they have a propensity to progress and interfere with daily activities. Therefore, the families of affected individuals are expected to identify this symptom early on and present the affected individual for screening or examination at a health care facility. © 2021, Associacao Arquivos de Neuro-Psiquiatria. BACKGROUND: Melasma has been suspected to be linked with levels of thyroid hormone. There is no study that explains the association between thyroid hormone levels with melasma severity. AIM: This study aims to find the discrepancies in the levels of thyroid hormone in varying severity of melasma using two different measurement techniques. METHODS: Subjects were chosen consecutively from the dermatology clinic at RSUPN Dr. Cipto Mangunkusumo hospital. Forty-eight patients participated in this study were categorized into mild melasma and moderate-severe melasma based on modified melasma area and severity index (mMASI) and Janus II measurement. RESULTS: Statistically, mMASI measurement showed no significant association between varying melasma severity with levels of thyroid stimulating hormone and free T4 (FT4), p = 0.375 and p = 0.208, respectively. The Janus II examination using polarized light modality has a weak positive correlation with the serum FT4 level (r = 0.3; p = 0.039). Weak correlation was also found between the two measurement strategies, Janus II and mMASI (r = 0.314; p = 0.03). Background. Multiple sclerosis (MS) is an autoimmune disease characterized by inflammation and demyelination of the central nervous system which often involves the optic nerve even though only 20% of the patients experience optic neuritis (ON). Objective. This study aims to compare the retinal structure and optic nerve function between patients with MS and healthy controls (HCs), evaluate optic nerve alterations in MS over 1-year follow-up, and analyze its correlations with disease duration, number of relapses, degree of disability, and different subtypes. Methods. This is a prospective cohort study involving 58 eyes of MS patients. Optic nerve function was evaluated with best-corrected visual acuity (BCVA), contrast sensitivity, and P100 latency, while the retinal structure was evaluated from the GCIPL and RNFL thickness measured with optical coherence tomography (OCT) and fundus photography. Results. The MS group had lower BCVA (p=0.001), contrast sensitivity (p<0.001), mean GCIPL thickness (p<0.001), and mean RNFL thickness (p<0.001) than HC. At 6 and 12 months of observations, GCIPL and RNFL (nasal quadrant) of MS patients decreased significantly (p=0.007 and p=0.004, respectively). Disease duration and the number of relapses correlated with delayed P100 latency (r = -0.61, p<0.001 and r = -0.46, p=0.02). GCIPL and RNFL in the SPMS subtype were thinner than in RRMS. Conclusions. The retinal structure and optic nerve function of MS patients are worse than those of normal individuals. GCIPL and RNFL thinning occurs at 6 and 12 months but do not correlate with disease duration, the number of relapses, and degree of disability. © 2021 Riwanti Estiasari et al. |
