BACKGROUND: Decreased Natrium iodide symporter (NIS) expression levels or diminished NIS targeting thyroid cancer cells’ plasma membrane leads to radioiodine-refractory disease. AIM: The aim of this study was to analyze the NIS expression in thyroid tumors. MATERIALS AND METHODS: The samples were thyroid tissues of patients who underwent surgery for a thyroid tumor. The tissues were processed for NIS protein expressions by immunohistochemistry (IHC) and Western blot (WB). Graves’ disease samples were used as positive controls. The samples were incubated without the primary antibody, and they were used as negative controls for IHC examination. Na+/K+ ATPase was a plasma membrane protein marker in the WB procedure. RESULTS: Twenty-nine samples were assessed for NIS protein. All of them showed the expression in the cytoplasm with intensity 1+ to 3+ with Allred score 3-8. Fourteen out of 29 cases (48.2%) showed NIS cytoplasm staining intensity ≥2+ consist of 10 papillary thyroid cancer (PTC), three follicular thyroid cancer, and one adenoma. Membrane staining was found in 2 samples of PTC (6.9%). Six samples (adenoma 1 sample, PTC 5 samples) showed NIS expression at membrane very weak (1+); they were considered as negative. NIS protein has several bands of ~ 80 kDa, ~ 62 kDa, and ~ 49 kDa. CONCLUSION: NIS expression in thyroid cancer mostly expresses in the cytoplasm instead of the membrane. NIS will play a functional role in the membrane to bring iodine across the membrane against the concentration. Ventricular septal rupture (VSR) is rare but a lethal complication of acute myocardial infarction (MI). Definite treatment requires the surgical closure of the VSR and coronary artery bypass grafting (CABG). However, the optimal timing for surgery is still controversial, particularly during the novel coronavirus disease 2019 (COVID-19) pandemic where medical procedures should be performed within the safest environment. Before surgery, a proper management in the intensive cardiovascular care unit is essential to maintain the stability of the hemodynamic profile related with VSR and determines the prognosis of the patient. We described a case of VSR complicating an anterior wall MI in a patient who admitted to our hospital during the COVID-19 pandemic that was treated successfully by surgical closure of the VSR and CABG. © 2021 BMJ Publishing Group. Introduction: Performance assessment of residents should be achieved with evaluation procedures, informed by measured and current educational standards. The present study aimed to develop, test, and evaluate a psychometric instrument for evaluating clinical practice performance among Dermatology and Venereology (DV) residents. Methods: This is a qualitative and quantitative study conducted from 2014 to 2016. A pilot instrument was developed by 10 expert examiners from five universities to rate four video-recorded clinical performance, previously evaluated as good and bad performance. The next step was the application of the instrument to evaluate the residents which was carried out by the faculty of DV at two Universities. Results: The instrument comprised 11 components. There was a statistically significant difference (p < 0.001) between good and bad performance. Cronbach’s alpha documented high overall reliability (α = 0.96) and good internal consistency (α = 0.90) for each component. The new instrument correctly evaluated 95.0% of poor performance. The implementation study showed that inter-rater reliability between evaluators range from low to high (correlation coefficient α =0.79, p < 0.001). Conclusion: The instrument is a reliable and valid instrument for assessing clinical practice performance of DV residents. More studies are required to evaluate the instrument in different situation. © 2021 TAPS. All rights reserved. |

Type 2 diabetes mellitus (T2DM) is one of the most common degenerative disorders. For therapeutic use, herbs are commonly used in Indonesia for T2DM treatment, one of them is (Cosmos caudatus) kenikir's leaves. In previous studies, kenikir's leaves have high antidiabetic and antioxidant activity. However, a comparison of antidiabetic activity from many extracts of kenikir's leave is remain unclear. This study will compare the antidiabetic and antioxidant properties of various kenikir's leave extract. Kenikir's leaves are extracted by maceration methods for three days using three different solvents: boiling water, 50% ethanol, dan ethanol 100%.Then, phenolic and flavonoid content will be measured, as well as antioxidant properties by DPPH radical scavenging activity assay, and antidiabetic properties by α-glucosidase inhibition assay, also LCMS/MS will be used to predict the compound from each extract. The result shows that 50% ethanol extract has highest phenolic and flavonoid content than others. It also has significantly higher antioxidant (p<0.05) and antidiabetic (p<0.05) properties than others. Meanwhile, LCMS/MS result of 50% ethanol extract predicts 6 chemical component, that quercetin is the most dominant compound. 50% ethanol extract of kenikir's leaves is superior from other extracts on phenolic and flavonoid content, antioxidant properties, and antidiabetic properties. © 2021 Phcogj.Com. The present study aimed to evaluate phytochemical and antioxidant activity (in vitro and in vivo) of Origanum vulgare (L.) ethanolic extract. The phytochemical test was assessed using the Clule method in ethanol, ethyl acetate, and hexane. In vitro evaluation of antioxidant activity was determined by radical scavenging assay using DPPH (2,2-diphenyl-1-picrylhydrazyl) as an artificial free radical activity. In vivo test was conducted to evaluate the effect of malondialdehyde (MDA) level in blood plasma during maximum physical activity treatment. In vivo test was done using 25 male Sprague Dawley rats in pre and post-test control group design. The phytochemical test of O. vulgare ethanol extract was showed some compounds, such as a flavonoid, alkaloid, triterpenoid/steroid, essential oil, and tannin, then in ethyl acetate and hexane. In vitro assay showed that O. vulgare extract has strong antioxidant activity with an IC50 value of 133.47 µg/mL. While in the in vivo test, the most effective dosage is 20 mg/200 gr B.W., represented by a significant decrease of MDA level (0.509 nmol/mL) before and after treatment. So, the ethanolic extract of clove has potency as an herbal antioxidant because of the low level of IC50 and can decrease the MDA level. © 2021 Phcogj.Com. Acne vulgaris is a chronic and self-limiting disorder of the pilosebaceous unit which is primarily seen in adolescents. Acne vulgaris presents as polymorphic lesions, consisting of comedones, papules, pustules, cysts, nodules, scarring, and dyspigmentation. Acneiform presentation of cutaneous lupus erythematosus (CLE) is extremely rare. The presentation of CLE is notoriously diverse and often mimics a broad range of unrelated skin disorders. We present a case by referring to American College of Rheumatology (ACR) criteria for systemic lupus erythematosus (SLE); our patient's conditions did not meet any of the clinical criteria of the Systemic Lupus International Collaborating Clinics (SLICC) for SLE. Subsequent to thorough history-taking, physical examination, and laboratory evaluations, the diagnosis of acne vulgaris was established, and a diagnosis of CLE was excluded. As acneiform presentation of CLE is rare, we here present a case which resembled both acne vulgaris and CLE. We describe our experience in establishing the diagnosis of severe acne vulgaris accompanied by scars in a 12-year-old boy with a malar rash and scars on his frontal and malar area who had initially been misdiagnosed as having CLE. This unusual case highlights the broad spectrum of adolescent acne and the importance of clinical identification of the disease so that unnecessary workups might be avoided. © 2021 |

Obesity is linked to more deaths worldwide. In obesity, there will be a dysregulation of growth signals such as tumorigenesis. Despite the fact that obesity is tend to progress to acute liver damage, not many study using 2-nitropropane (2NP) as a hepatoxicity agent are undertaken especially in obese mice. This study aimed to determine the regime of 2NP that causes acute liver damage. This is an experimental research using a post-test control design group only, with 3 groups of mice ie O1 (obesity), O2+2-NP 1x (induced by 2NP 100 mg/kg BW once), and O2+2-NP 2x (induced by 2NP 100 mg/kg BW twice). At 10 weeks, rats were sacrificed and 100 mg liver tissue were collected for MDA, GSH, MnSOD and CAT enzymes analysis. Analysis statistics were performed by SPSS by one-way Anova and post hoc Tukey. MDA levels of mice were found to be increased in 2NP group than control (3.768 ± 0.407 nmol/ mg) (p < 0,01). Liver GSH, MnSOD and CAT levels of both single injection 2-NP and repeated injection 2-NP groups decreased compared to those of controls (p<0,01). Purpose: The prevalence of rickets is increasing worldwide in association with an increase in vitamin D deficiency. This study aimed to investigate the vitamin D profile of healthy school-aged children in a sun-rich country and its contributing factors. Methods: This cross-sectional study was conducted in 120 healthy children from 7–12 years of age who live in Jakarta, Indonesia. Their demographic status, sun exposure duration time, and lifestyle were recorded using a structured questionnaire. Serum calcium, phosphate, bone-alkaline phosphatase (B-ALP), and 25-hydroxy vitamin D (25(OH)2D3) levels were measured. The participants were categorized into vitamin D sufficient and non-vitamin D sufficient groups, and we analyzed variables that contributed to the 25(OH)2D3 level. Results: Of the participants, 73 (60.8%) were vitamin D sufficient, 45 (37.5%) were vitamin D insufficient, and 2 (1.7%) were vitamin D deficient. Sex, age, body mass index, Fitzpatrick skin type, daily milk intake, and clothing type were not different between the vitamin D sufficient and non-vitamin D sufficient groups. There were no differences in serum calcium, phosphate, and B-ALP between the 2 groups. Sun exposure time was significantly longer in the vitamin D sufficient group compared with that in the non-vitamin D sufficient group (511.4 min/wk vs. 318.7 min/wk, P=0.004), and this effect remained consistent on multivariate analysis after adjustment for covariates (adjusted odds ratio, 1.002; 95% confidence interval, 1.000–1.003). More participants in the vitamin D sufficient group did not use sunscreen (59 vs. 27, P=0.02), but this finding was inconsistent with our multivariate analysis. Conclusion: Despite year-round sun exposure, approximately 1 in 3 primary school-aged children had insufficient vitamin D level. Titanium (Ti) has been used in metallic implants since the 1950s due to various biocompatible and mechanical properties. However, due to its high Young’s modulus, it has been modified over the years in order to produce a better biomaterial. Tantalum (Ta) has recently emerged as a new potential biomaterial for bone and dental implants. It has been reported to have better corrosion resistance and osteo-regenerative properties as compared to Ti alloys which are most widely used in the bone-implant industry. Currently, Tantalum cannot be widely used yet due to its limited availability, high melting point, and high-cost production. This review paper discusses various manufacturing methods of Tantalum alloys, including conventional and additive manufacturing and also discusses their drawbacks and shortcomings. Recent research includes surface modification of various metals using Tantalum coatings in order to combine bulk material properties of different materials and the porous surface properties of Tantalum. Design modification also plays a crucial role in controlling bulk properties. The porous design does provide a lower density, wider surface area, and more immense specific strength. In addition to improved mechanical properties, a porous design could also escalate the material's biological and permeability properties. With current advancement in additive manufacturing technology, difficulties in processing Tantalum could be resolved. Therefore, Tantalum should be considered as a serious candidate material for future bone and dental implants. © 2021 Trans Tech Publications Ltd, Switzerland. |
